Both Murschell and Sayre competed in last year's tournament in Las Vegas, and were drawn together for the opening fixtures of the 2018 North American Championship at the Mandalay Bay Casino & Resort.
Canada's Murschell sealed his place in the North American Championship by reaching the semi-finals of Wednesday's Qualifier Four, while American ace Sayre was the Qualifier One winner on Tuesday.
John Norman Jnr, the Qualifier Two winner, has drawn fellow Canadian David Cameron, while Qualifier Three winner Jeff Smith will play Ross Snook in another all-Canadian tie.
Youngster Danny Lauby will take on Joseph Huffman in a clash of two big-stage debutants in the other tie, with the pair also having faced off in Wednesday's Qualifier Four final.
The North American Championship, won last year by Willard Bruguier, will play to a conclusion on Thursday, with a $10,000 first prize for the eventual winner accompanied by a spot in the William Hill World Darts Championship.
The draw for the William Hill US Darts Masters will be made between the semi-finals and final of the North American Championship, ahead of the main World Series of Darts event on Friday and Saturday.
